Proposed Analysis: | Dear ADNI team, I am clinician-scientist with a special interest in the characterisation of white matter injury in Normal Pressure Hydrocephalus seeking to obtain ADNI data as a comparator arm for NPH analyses. I currently run multiple studies involving the use of multi-modal imaging and fluid biomarkers in NPH, including overlay with neurodegenerative conditions and dementia. I hold active grants from the following funding bodies - National Medical Research Council Transition Award, Centre grants and a Khoo Pilot Award. My main focus is on the use of diffusion tensor imaging for the examination of white matter injury, cross-correlations with multi-modal MR data, blood and CSF biomarkers. I seek to understand the correlates within a purer AD subtype in order to separate overlay pathologies within the NPH spectrum. My proposed analyses for the ADNI data include structural, DTI post-processing and ROI/ tractography analyses as well as the use of biomarker data to understand thresholds for AD pathology vs. NPH overlay. In addition to NPH, I also plan to recruit an elderly cohort of minor traumatic brain injury (TBI) to undergo imaging locally, in accordance with the published ADNI MR protocols, in order to have appropriate age-matched comparators for frail elderly with fall risk. The overall trajectory of my research programme is to understand and characterise the spectrum of NPH from early to late subtypes. As NPH is rare in the population, access to the ADNI database would allow me to have more robust comparisons to known and validated imaging biomarkers of cognitive burden.
