Proposed Analysis: We are requesting access to the ADNI dataset (see full proposal for data requested) in order to examine hyperconnectivity hypotheses in resting fMRI data. We propose using a cohort design representing distinct age groups over the compacted life span of ADNI participants. We will leverage the ADNI dataset to address three Aims. First, we will document the most reliable connections observable across distinct aging cohorts with the goal of determining if there are age-related maturational processes that alter the variability of network connectivity and the location/role of hubs. To achieve this goal, we will track the variance in the most reliable connections across the selected cohorts (see Abraham et al., 2017, Neuroimage). Second, we aim to determine if hyperconnectivity can be reliably observed across cohorts in the context of changing plasticity with age (including factors such as estimated cognitive reserve, sex, and structural brain volume). Related to this goal, we anticipate that brain injury may elicit hyperconnectivity as a compensatory response, but the time line and chronicity of hyperconnectivity after injury is uncertain and it is unclear if this process mirrors what is observed in MCI. So, as part of this second goal, we will identify common sites for hyperconnectivity in brain injured cohorts, how hyperconnectivity is sustained based upon time since injury/diagnosis, and the potential for later connectivity loss with age and/or onset of MCI (see 6,10) (see Figure 1 in proposal). Finally, we aim to determine if hyperconnectivity in critical network hubs in each of the cohorts predicts behavioral outcome including non-linear relationships. In order to address these questions, we are requesting data for individuals in the ADNI dataset who fit the following categories: 1) healthy aging, 2) prodromal AD/MCI, and 3) traumatic brain injury. Appendices I and II in the proposal outline the specific data requested.
