Proposed Analysis: I am a colleague of Brian Ott at Brown. Our original application was to study the blood-brain barrier, and our intention was to assess for indicators of BBB function in preclinical AD, however the biomarkers we were interested in using were not available in the proteomics data set. We then shifted focus to biomarkers vascular endothelial dysfunction in AD, the results of which we presented in a poster at the 2018 AAIC conference. The results of that study showed that, in a cross-sectional analysis, elevated levels of vascular cell adhesion molecule-1 (VCAM-1) predicted AD diagnosis and functional decline (as measured by CDR sum of boxes) in ADNI 1 participants, but did not correlate with CSF amyloid/tau or ApoE. Moving forward, we intend to continue with this line of inquiry by expanding our analysis to include inflammatory biomarkers (peripheral and CSF), in addition to performing a similar, longitudinal study.
