Proposed Analysis: | Emerging studies have suggested that tumor necrosis factor-alpha (TNF-α) is implicated in the pathogenesis of Alzheimer’s disease (AD), and that cerebral glucose hypometabolism is a key feature of AD. However, the association of CSF TNF-α levels with change in cerebral glucose metabolism, as measured by FDG-PET, has not been studied among non-demented older people. We aimed to examine the cross-sectional and longitudinal associations of CSF TNF- α with global cognition, verbal memory, and brain glucose metabolism (as assessed by FDG-PET) among non-demented older people from Alzheimer’s Disease Neuroimaging Initiative (ADNI) study. |

Proposed Analysis: | We aimed to investigate the association of CSF TNFα and its related receptors with the risk of developing dementia among MCI subjects. First, the Youden-based cutpoints rather than the medians were applied to binarize CSF TNFα and its related receptors. Second, several Cox proportional hazards regression models were performed to evaluate the associations of CSF TNFα and its related receptors with conversion to dementia with adjustment of several covariates. |
