Proposed Analysis: Neuroaxonal damage is a shared and early feature of neurodegenerative diseases, including Alzheimer disease. Neurofilament light chain (NfL) levels in blood have recently emerged a sensitive preclinical maker of neuroaxonal damage. Nevertheless, little is known about the genetic and epigenetic determinants of NfL levels. Therefore, we aim to perform genome- and epigenome-wide association studies of blood NfL levels in a large-scale population-based cohort study. We aim to use (genomic and epigenetic) data from ADNI for replication and external validation of our findings in a fully independent cohort.
