Proposed Analysis: | Stress and stress-related biomarkers are correlated with cognitive decline and Alzheimer’s Disease (AD) pathogenesis. Thus, these parameters may serve as preclinical markers for AD. We plan to investigate the relationship among stress-associated biomarkers and single-nucleotide polymorphisms (SNPs) in ADNI1 study participants (n=818). Specifically, we are requesting: • 39 stress- and cognition-association SNPs (37 from the Illumina Human 610-Quad bead chip and 2 APOE genes), and biomarkers of cumulative stress (body mass, systolic and diastolic blood pressure, pulse, and homocystine) measured at both baseline and 24-month follow up. We plan to use structural equation modeling to estimate relations among these biological measures and various cognitive measures (see immediately below). • Scores from the following cognitive assessments (measured at baseline and 24-month follow up): American National Adult Reading Tests, Mini Mental Status Examination, Logical Memory 1 and 2, Everyday Cognition, Montreal Cognitive Assessment, Digit Span, Category Fluency, Category Fluency Animals, Trails Test, Digit Symbol, Boston Naming Test, Auditory Verbal Learning Test, Clock Drawing, ADAS-cog, ADAS-cog13, Clinical Dementia Rating Scale. • Cerebrospinal fluid Tau, AB1-42 and P-tau181P data to compare levels of these AD-associated proteins with stress-associated biomarkers. • If plasma or serum exists for the ADNI1 cohort at baseline, we would like to request access to aliquots (325 ul) for analysis of circulating cortisol and catecholamines, two important stress-related hormones. Based on a search of the ADNI literature database, this is a novel proposal and its completion will provide much needed information on novel AD biomarkers. |
