Genetics & Related – Omics

The data made available through the genetics core includes a wide variety of genetic and other -omics data. The data sets hosted on the LONI IDA include information on the following:

Genomic Data

APOE
genotype

APOE genotypes have been determined using evolving methods over the past two decades and are available in the ADNI clinical database and can also be downloaded here.

Whole genome sequencing

ADNI WGS has been performed in three phases. The first phase is available on the LONI IDA website. The first two ADNI WGS datasets are currently available through the NIAGADS Data Sharing Site which is the recommended method to obtain cleaned, integrated and jointly called data through the NIA ADSP. A third phase of WGS is expected as part of ADSP Release 5 later in 2024.

Genome-wide association studies

Genome-Wide Association Studies: GWAS is available for the ADNI-1/GO/2/3 phases. GWAS will be generated for ADNI4 participants at periodic intervals and posted here.

Epigenetics (DNA methylation, etc.)

Longitudinal Illumina EPIC arrays and telomere length measurements are available for selected ADNI participants. Additional ADNI DNA methylation data is planned through CLEAR-AD

Transcriptome (RNA, miRNA)

Gene expression profiles are available for selected ADNI-GO/2 participants. Large-scale longitudinal RNA-sequencing data are being generated by CLEAR-AD and will be available here and on the Sage AD Knowledge Portal (expected beginning in late 2024; see CLEAR-AD for status updates). In mid-2024 micro RNA sequencing on baseline ADNI plasma samples was generated by a collaborative project led by Ivana Delalle, Andre Fischer and colleagues (Alzheimer’s & Dementia in press).

Multiple proteomics platforms have been run on ADNI plasma and CSF samples over more than a decade. More information can be found under the ADNI Biomarker Core. Selected analyses have been conducted by investigators from Emory, Washington University, and the FNIH Biomarker Consortium among other groups. Additional cross- referenced materials can be found here in the future.

metabolomics & lipidomics

Multiple data sets have been generated from ADNI 1/GO/2 serum by the AD Metabolomics Consortium (ADMC) led by Rima Kaddurah-Douk in coordination with the Genetics Core. Data has been generated for as many as 10 platforms. Many are unblinded and posted to LONI and others are in the process of QC and release.
