Proposed Analysis: | Mounting evidence of the role of cerebral small vessel disease (SVD) in Alzheimer's disease (AD) pathology highlights the need to understand its role in AD pathogenesis. While some have examined SVD in relation to the longitudinal pathological accumulation of amyloid and tau, the mechanisms involved remains uncertain. To extend this field of inquiry, we propose examine AD pathology in relation to different types of spatial distributions of SVD, which have been shown to be caused by distinct pathologies. To do so, we will quantify the degree of SVD burden on structural MR scans, and analyse these in relation to cerebral amyloid and tau burden as measured on PET imaging. To expand on related areas of research conducted in ADNI, we will focus on (1) the regional distribution and type of SVD, (2) the interaction of these pathologies on clinical outcomes such as cognition, and (3) whether these processes differ across the lifespan. |
