Proposed Analysis: | We wish to explore the effect of Type 2 Diabetes Mellitus on brain biomarkers The broad aim is to compare patterns of brain metabolism (FDG-PET), amyloid uptake (PiB), and structural MRI measures between patients with Type 2 Diabetes (not cognitive impaired, MCI, Dementia), those with MCI/Dementia but no Type 2 Diabetes, and Healthy Controls. The analysis will stratified by severity of cognitive impairment (eg CDR)and glucose control (plasma fasting glucose, Hba1c). Additionally, if CSF biomarkers are available, they will add substantially to the comparisons being made. The hypothesis is that DM will be associated with these biomarkers of neurodenerative disease (global and regional analyses), and this will be greater with increasing severity of cognitive impairment. This planned to be a voxel-based comparison using established Statistical Parametric Mapping (SPm) methodology, as well as other novel approaches to regional analyses. We are proposing this analysis based on our MRI structural findings in a sample of community-dwelling people with Type 2 Diabetes Mellitus. Our group has significant experience in brain imaging analyses using established and novel voxel-based approaches, particularly in large datasets. |
