Proposed Analysis: This study will examine head injury exposure on brain indicators of accelerated aging. The specific aims are as follows: i) to characterize the potential factors that may contribute to accelerated aging by examining the relationship between a history of mTBI and SCI on cortical/subcortical thickness/volumes in disease-vulnerable regions; ii) to assess the effects of age of first injury and head injury exposure on measures of cortical and subcortical thickness/volume in healthy adults; and iii) to examine the indirect influence of head injury history on the relationship between neuroimaging metrics of brain health and cognitive status (i.e., based on neuropsychological test outcomes). The relationship between performance on cognitive tasks will also be compared in those with a history of mTBIs/SCIs, across the lifespan. For the present study, the hypotheses are as follows: I. Individuals with an increased number of previous head injuries (i.e., mTBI and/or sub-concussive impacts) will demonstrate greater cortical thinning and smaller subcortical volumes in AD-vulnerable regions than those with fewer or no head injuries. II. There will be an age by head injury exposure interaction, with older adults demonstrating greater differences in cortical and subcortical neuroimaging metrics than age-matched controls. III. Age at first injury will be negatively associated with cortical thickness and subcortical volumes. IV. i) Reductions in cortical thickness/subcortical volumes will be associated with poorer performance on a delayed memory task and ii) concussion history will indirectly influence the relationship between cortical thickness/subcortical volumes and delayed memory performance.
