Proposed Analysis: To evaluate the correlation between CSF AD tau and Aβ42 levels with regional FDG PET, in normal controls, MCI and AD subjects. While the changes in tau and Aβ42 CSF levels and FDG PET patterns are validated biomarkers of AD pathology, their relationship remains unclear. The results available in the current literature are inconsistent: while Vukotic et al. (Dement Geriatr Cogn Disord 2009; 27:474) find a correlation between Aβ42 levels and FDG, but no correlation between tau levels and FDG, in 32 patients with mild to moderate AD, the study by Petrie et al (Arch Neurol 2009; 66:632) finds an negative association between tau and FDG in 20 healthy volunteers and a positive association between Aβ42 levels and FDG. The ADNI database will provide a definite answer to this question in normal subject, patients with early MCI, MCI and AD. In addition, the analysis of longitudinal data will provide additional information about the evolution of glucose metabolism and CSF marker changes.
