Proposed Analysis: We plan to test whether cognitively normal older adults (age greater than 60) with high levels of Alzheimer's (AD) related pathology, such as tau and beta-amyloid, have better protection against gray matter atrophy and cognitive decline if they have certain genetic polymorphisms related to dopamine function (COMT, DRD2 genes). Specifically, we will be analyzing longitudinal 3T structural MRI scans to measure gray matter atrophy over time, and testing whether certain genetic polymorphisms are related to better or worse preservation of gray matter in individuals with varying levels of AD-related pathology. Further, we will test whether having certain polymorphisms is related to better executive function and memory for individuals who have large amounts of atrophy.
