Proposed Analysis: Dr. Boerwinkle’s group has been involved in all aspects of the ADSP including study design, data gathering and data analysis. Permissions are necessary for both the array genotype, DNA sequence and all available phenotype data. This availability and ensuing data access will be used for data processing and data analysis to identify novel AD risk raising and protective loci.

Proposed Analysis: We analyze ADNI genetic and phenotypic data as part of the Alzheimer’s Disease Sequencing Project (ADSP) to identify genes and pathways influencing risk of AD. Specifically, ADNI whole genome sequence (WGS) data was processed in conjunction with other ADSP data for calling of single nucleotide polymorphism and structural variants and is an integral part of the full ADSP dataset made available on NIAGADS. We are performing WGS analyses of the entire ADSP dataset to identify variants associated with AD. In the analysis of the first 5K samples (including ADNI), we have incorporated AD-relevant functional annotations in our WGS analyses and identified 2 rare variants on chromosome 17. Analyses on an expanded datasets is ongoing. ADNI is also part of this dataset.
