Proposed Analysis: | We have spent the last years molecularly profiling different regions of the brain in the mouse, and have come up with a shortlist of a few genes that are enriched in the layer II of the entorhinal cortex, and CA1, that are the first ones which accumulate NFTs in AD. Since Abeta is not particularly appearing around specific neuron types, but is more widespread that NFTs, we think that these regions are more prone than other regions to forming NFT in response to Abeta deposition. We would like to search for potential protective SNPs in our ECII and CA1 enriched genes that could prevent people from converting to AD in the presence of high levels of ABeta. We will study SNP genotyping data in conjunction with Amyloid imaging and clinical diagnostics to pinpoint potential associations around our candidate genes. |
