Proposed Analysis: | One of the primary purposes of neuroimaging of tau in AD subjects is to identify tau-positive scans. Published approaches, such as SUVR of 'composite' regions did not discriminate scans of AD subjects from age-matched control subjects (OC) as accurately as one could wish for. We introduced the tau-positive fraction (TPF) which is defined in individual regions as the frequency of voxels that exceed region-specific threshold (= mean + 2.5 of OC) on SUVR images. This variable successfully discriminated AD from OC subjects (specificity: 94% with ‘composite’ TPF; 100% with maximal TPF), compared to ‘composite’ SUVR (82%) in a small cohort (11 AD and 5 OC subjects) using [18F]RO-948 (Wong et al., JNMMI 2018). We would like to test if TPF could improve identification of tau-positive scans with AV-1451 using the ADNI dataset. |
