Proposed Analysis: The objective of this study is to analyze genomic data from individuals with prostate/breast cancer (the most common cancer type in aging populations) as compared to those with Alzheimer’s disease. In doing so, we hope to hone in on novel genomic regions that are potentially conferring risk for one condition rather than the other. Individual genome-wide SNP data collected by the Breast and Prostate Cancer Cohort Consortium (BPC3) and ADNI will be obtained from dbGaP/LONI, respectively, and stored on a local, password protected server within a mag-card secured space at the UNTHSC. A subset of individuals from the BPC3 cohort will be selected who are age-, gender-, and population-matched to the cases from the ADNI cohort for Alzheimer’s disease. Both ADNI and BPC3 were genotyped using the Illumina arrays which includes 550,000+ SNP markers. Genotype calling and principle component analysis (to account for population substructure) will be performed prior to association testing using a logistic regression model in PLINK (Purcell et al.)and Bayesian Mulitnomial regression models in Trinculo (Jostins et al., 2016). Genome-wide association signals, in theory, will indicate regions of interest which are implicated in the inverse relationship between AD and cancer. In accordance with best practices for data sharing, data will be obtained and used under strict adhesion to the following: • Data will only be used for the purposes specified within this proposal and will remain de-identified; • Data will not be sold or shared with anyone other than the investigators listed herein; • If requested by the primary owners/issuers of the data, periodical reports, requests for renewal, and close out of projects will be honored. • Any violation of the NIH’s Genomic Data Sharing Policy will be reported as soon as it is discovered. • A listing and summary of this project may be made publically available. • The contributing investigator(s) who conducted the original study will be acknowledged as specified in the Data Use Agreement. This project is preliminary and will be executed under the sole control of the PI. Collaborators on previous similar GWAS projects will likely be lending expertise and advice, but will not be directly involved with the data analysis.
