Proposed Analysis: The various neuropsychiatric symptoms (NPS) have shown to predict higher risk of dementia, and have been postulated to be prodromal symptoms of dementia. However, it is not clear whether the various NPS are related to similar neuropathology or represent distinct neuropathologies. This study sought to identify the distinct brain regions that are affected by the various NPS, using multivariate analysis to examine the association between various NPS (as identified on Neuropsychiatric Inventory) and individual brain volume (as captured on MRI).

Proposed Analysis: Much research efforts and funds have been invested in the last decades to find a disease modifying drug for Alzheimer’s dementia (AD) (the most common subtype which accounted for 60-80% of all dementia cases)("2016 Alzheimer's disease facts and figures," 2016). However, 99.6% of these research trials failed to find a useful drug which may potentially be used for AD (Cummings, Morstorf, & Zhong, 2014). Many reasons have been posited for such high failure rate. One reason may be related to our still limited understanding of the pathogenesis of AD. This study seeks to utilize the genetic data in ADNI (including DNA, DNA methylation and Gene expression data) to examine their associations with phenotypes that are related to AD (including clinical diagnosis of dementia, neuroimaging biomarkers and CSF biomarkers), with the aim to improve our understanding on the pathogenesis and biological pathways of AD.
