Proposed Analysis: Alzheimer’s disease (AD) and Parkinson's disease (PD) are neurodegenerative disorders and brainchanges can be investigated non-invasively using Magnetic Resonance Imaging (MRI). Gyrificationof the brain’s cortex takes place during brain development and it is among the fundamental featuresof primate neuroanatomy. The mechanisms of brain folding are not fullyunderstood, but the process is preserved among human and non-humanprimates. The complexity of brain gyrification reflects a combination of many factors, includinggenes, environment and pathological changes. We recently report theheritability of sulcal phenotypes on 4 large independent cohorts. We propose to perform an in-depth investigation of the cortical folds using structural brain MRIs fromavailable AD and PD datasets such as ADNI (http://adni.loni.usc.edu/) and PPMI (http://www.ppmi-info.org/). Major expected outcome:1) we will define a robust model able to predict earlier the progression of the disease; 2) using genetic data, fully available for the datasets described here, we will define the genetic riskscores for AD and PD and we will give new insights on the pathogenesis of neurodegenerativediseases.
