Proposed Analysis: In recent years, the relationship between uric acid and Alzheimer's disease (AD) has been widely concerned. Epidemiological studies have found that serum uric acid (UA) is lower in AD patients than normal elderly people, supporting that UA plays a protective role in the cognitive decline and dementia. Our results suggested that UA provoked cognitive function improving in 5XFAD mice, which was related to the endocytosis and degradation of Aβ oligomers by microglia. Now we hope that we can get more clinical information from ADNI database to analyze the correlation between serum or CSF UA levels and cognitive functions. In addition, we want to investigate the appropriate dose of UA administration to increase CSF uric acid levels through comparing the serum and CSF UA levels in normal individuals and AD patients. Finally, we want to determine the correlations between uric acid and AD biomarkers such as Aβ and tau. Currently, we have basically completed relevant experiments on AD mice and BV2 cells and we are exploring mechanism researches on primary microglia. We hope to explore the protective role of UA in the occurrence and development of AD, and as a new approach for AD treatment.
