Proposed Analysis: We are primarily interested in studying changes to the brains functional architecture in Alzheimer’s and how this varies with severity of disease and cognitive decline. We propose to use the resting state fMRI data to identify the major intrinsic connectivity networks of the brain and to study how within and between network connectivity varies with neuropsychological and cognitive assessments in these patients. We are primarily interested in network relationships with the following characteristics: Alzheimer’s Disease Assessment Scale (ADAS), Clinical Dementia Rating (CDR), Cognitive Battery results, Montreal Cognitive Assessment (MoCA). We will first apply group spatial ICA to the fMRI data using FSL Melodic and then use subsequent dual_regression analysis to identify individual subject maps and timecourses of network activity. Randomise permutation testing will be used to study variations in network strength and spatial extent between subjects with lowest and highest characteristics. In addition to the cross sectional design we will also explore linear correlations across the whole group. Furthermore network modelling will be applied using FSLnets to study how between-network connectivity differs between the patients with the lowest and highest Alzheimer’s and Dementia ratings, and also between those with the most and the least cognitive decline.
