Proposed Analysis: The intended analysis aims to delve into the intricate interplay between longitudinal shifts in amyloid-PET and tau-PET burdens, cognitive decline, and potentially cortical atrophy. A primary emphasis will be placed on investigating the interrelationships between the varying rates of marker alteration, alongside possible modifiers like age, gender, genetics, and hypertension. The forthcoming phases of the study will unfold as follows: 1. Ascertain the specific Centiloid value(s) that signify a discernible shift in trajectory. 2. Identify the point(s) marking a change in the trajectory of flortaucipir binding. These values are compared and examined for discrepancies between the risk factors. Depending on the result, they can be included in subsequent analyses. 3. Disqualify participants lacking a minimum of two amyloid PET scans and two tau PET scans. 4. Harmonize amyloid and tau PET scans, ensuring a maximum temporal separation of one year. If unattainable, the participant will be omitted. 5. Apply linear mixed-effects models to determine individual slopes for each participant that determine amyloid and tau increases and cognitive decline. 6. Use structural equation modeling to assess the predictive power of baseline data and slopes of amyloid and tau in predicting cognitive decline (and, where possible, brain atrophy). 7. Probe the potential influence of moderating factors on these interconnected associations. By leveraging the potential of multivariate longitudinal data, the overarching purpose of this analysis is to discern the prime determinants that optimize forecasts of impending cognitive deterioration and elucidate the variables contributing to the intricate progression of Alzheimer's disease.
