Proposed Analysis: | Alzheimer's disease (AD) is a complex multifactorial neurodegenerative disorder. The greatest known risk factor for AD is advancing age. In this research study, we propose to perform association studies between age and molecular traits including clinical biomarkers, proteomics and metabolomics 1) within each gender group; and 2) within each disease group, i.e., NC (Normal Control), MCI (Mild Cognitive Impairment), and AD (Alzheimer's disease); 3) then compare the differences and similarities of the molecular patterns across gender and disease groups. The molecular genomic data proposed to access will include proteomics and metabolomics data measured in serum. To increase the power, we will also apply an interaction model for testing 1) the effects of age and sex; 2) and the effects of age and disease status on the molecular changes. Finally, the genotyping data including APOE genotyping will also be included to test the genetic determinants of the identified age-associated molecular patterns. We expect our study will identify age-related molecular signatures which can provide insights into the biology functions of advancing age in the pathogenesis and progression of AD. |
