Proposed Analysis: | For every participant identified as amyloid-positive (CSF Aβ1-42 ≤192pg/ml), I will extract across multiple time points: a)Medial temporal lobe (MTL) gray matter volumes b)Fractional anisotropy and mean diffusivity of fornix c)Auditory Verbal Learning Test and d)Logical memory Then, I will carry out the following analyses: 1.Cross-lagged panel analyses of MTL atrophy and fornix diffusion indices; specifically structural equation models (SEM) in which baseline MTL atrophy predicts subsequent fornix diffusion indices, while controlling for baseline fornix diffusion indices and other covariates, as well as the reverse – fornix diffusion indices predicting subsequent MTL atrophy, while controlling for baseline MTL volumes and other covariates 2.Also using SEM, I will examine the additional variance explained by fornix diffusion indices on top of MTL atrophy, and vice-versa, in predicting subsequent memory (latent factor of Auditory Verbal Learning Test and Logical memory), while controlling for baseline memory and other covariates |
