Proposed Analysis: | On behalf of the Neural Vitality Lab at University of Massachusetts Amherst, I would like to request access to ADNI data in order to analyze a subset of structural data encompassing emotion regulation networks in Ab +/- individuals that are clinically normal. Our research group recently wrote a position paper on anatomical and structural biomarkers of emotion dysregulation in the AD prodrome, identifying weakened network connectivity between frontal control regions (ACC, PFC) and limbic regions, as well as structural atrophy associated with higher rates of conversion from amnestic MCI to AD. Affect and neuropsychiatric health is typically a secondary or tertiary outcome interest in clinical trials but may improve diagnostic specificity in conversion along the AD trajectory. However, little literature (if any) exists on the functional and structural changes of emotion regulation regions early, in preclinical AD groups. Our group proposes to analyze differential (Ab +/-) resting state functional connectivity and structural integrity of limbic and frontal control regions associated with emotion regulation strategies such as reappraisal, which is particularly cognitively taxing for older adults. We hope to identify the earliest pathological changes associated with emotion regulation structures, and the implication of this for neuropsychiatric health, impairment, and conversion to clinical states. |
