We are mainly interested in MRI and PEN image data for research purposes such as drug discovery, disease prediction, and knowledge resourcing.

Proposed Analysis: Type 2 diabetes mellitus (T2DM) is a known risk factor for Alzheimer’s disease (AD). Studies show that metformin, a commonly used antidiabetic agent, is associated with lower incidence of AD and improved cognitive function. However, the underlying mechanisms of metformin improving brain function in AD remain largely unknown. We used Alzheimer’s Disease Neuroimaging Initiative database of 1740 participants (310 AD, 786 mild cognitive impairment, 473 normal) to examine the effect of metformin on brain volume. Several analyses have been performed. First, we used association rule mining of medical history of ADNI participants to investigate the comorbidities of AD, we found 23 comorbidities, among which hyperlipidemia, hypertension and diabetes ranked at top. Second, we used multivariate regression to analyze the association of metformin usage with brain volume in baseline. We found that metformin usage is significantly associated with increased whole brain volume (volume increase: 39 cm3; CI: 0.5 - 77.5, p = 0.047). Furthermore, metformin significantly targets a brain sub-region - rostral middle temporal cortex (volume increase: 2.55 cm3; CI: 0.94 - 4.16; p = 0.0019). We plan to leverage the longitudinal information in ADNI to further investigate the effect of metformin usage on change rate of brain volume.
