Proposed Analysis: Title: Racial differences in the impact of APOE and demographics on amyloid positivity Alzheimer’s disease (AD) is a global problem affecting rapidly ageing countries and areas such as US, Australia, Europe and East Asia. Amyloid has been suggested as one of the most important causes of AD. Therefore drugs targeting amyloid are in active developments and clinical trials worldwide. To make this global effort more effective, it is important to understand the mechanisms related to amyloid using data from multiple countries with different racial backgrounds. Jansen et al. (2015, JAMA) showed the impact of APOE and demographics on the prevalence of amyloid positivity using data from multiple countries. However racial differences in these factors are largely unknown. In this project, we plan to investigate the racial differences in the effects of APOE genotyping and demographics on amyloid positivity. We will do this by gaining access to amyloid cohorts from different countries with diverse racial backgrounds, e.g. ADNI (US), AIBL (Australia), DPUK/EMIF (UK/Europe) and Korea amyloid study (East Asia). Data storage, analysis and access will be strictly managed within a secure environment such as university firewall protection or special facilities provided by the Dementias Platform UK (DPUK).
