Proposed Analysis: | We plan to investigate the structural differences in the cerebral cortex between patients with AD and controls as indexed through measures of cortical thickness and cortical surface area. T1w MRI scans will be run through the FreeSurfer v6.0.0 and resulting cortical measures will be extracted through cortical reconstruction pipeline. Difference in thickness and surface area for each of the 34 regions of the Desikan-Killiany atlas will be analyzed using multiple linear regressions with age and sex as fixed effects. The profiles of beta-coefficients from these models will then be further investigated for underlying neurobiology as they index the "difference" of thickness and area between AD and controls. Resources from the Allen Human brain atlas will be used to correlated gene expression profiles with these difference profiles mentioned above. This methodology may help us understand the underlying neurobiology of structural changes within the cerebral cortex in AD. |
