Proposed Analysis: | Amyloid deposition and medial temporal lobe (MTL) atrophy are both well-known markers of Alzheimer’s disease. However, it is unclear how the two are linked in earlier stages of the disease. We hypothesized that the association between regions with relatively high 18F-florbetaben (amyloid-β) uptake and medial temporal lobe atrophy might be via disruption of the white matter of the hippocampal cingulum bundle. Preliminary results from our own MCI cohort (n = 24) suggest that 18F-florbetaben uptake was indeed associated with FA/MD of the hippocampal cingulum bundle but not with the superior part of the cingulum bundle. In turn, the FA/MD of the hippocampal cingulum bundle was associated with MTL volume. Furthermore, there was no direct association between 18F-florbetaben uptake and MTL volume. We would like to use the ADNI sample to be able to independently validate our hypothesis in a larger sample of patients with MCI with comparable high-quality MRI and PET data. |
