Proposed Analysis: A longitudinal study combine genetic variant, PET, MRI together to detect new biomarker to predict cortical atrophy. Trace back AD patients’ changes from Normal to MCI to AD, ROIs could be identified that are significantly related to AD progression and at the same time differentiable from control at normal stage. Then run anunbiased estimation of genetic variants that predict the ROIs with regression, then compare against genetic variants that be reported to be relevant to beta amyloid and tau disposition. In this way, we could construct a genetic-pathology-morphology chain for better understanding the mechanism undergoing AD progression.
