Proposed Analysis: The principal goal of this study is to identify novel associations between genomic data and phenotypic data to better understand and characterize dementia. Specifically, using both supervised and unsupervised AI techniques on the phenotypic data, we plan to identify endotypes of dementia. We anticipate that these dementia endotypes stratify individuals based on their different trajectory of disease progression, linked to specific aetiologies. Leveraging our expertise in integrative clinical and genomic analysis, we then will identify genetic signatures (both rare variants and polygenic scores from common variants) for each of these endotypes. Integrating these genomic and phenotypic signatures could enable stratification of individuals for personalized and preventive medicine, and identification of new targets for intervention.
