Proposed Analysis: Title: Fibrillar amyloid deposition and whole-brain structural connectivity analysis in the ADNI population. Purpose: To analyze the relationship between fibrillar amyloid deposition and whole-brain structural connectivity in the ADNI population. Background: The primary hypothesis for the project is that whole-brain structural connectivity in the ADNI population, as measured by diffusion tensor metrics calculated from diffusion tensor images, will change with increasing beta amyloid deposition, as measured by amyloid PET imaging. Subject inclusion / exclusion criteria: All subjects in the ADNI2 population who have anatomical, DTI, and F-AV-45 PET imaging available for each time point analyzed (e.g., baseline and follow-up). Analytic Plan: The DTI scans will be used for generation of structural connectivity graphs. The F-AV-45 PET images will be used for assessment of fibrillar amyloid deposition. Appropriate statistical models will be used for the assessment of significant changes in structural connectivity with respect to fibrillar amyloid deposition in both cross-sectional and longitudinal analysis between and within subsets of the ADNI population (i.e., cognitively normal, early MCI, late MCI, mild AD).
