Proposed Analysis: In a recent publication of our group (Ran et al. Brain Behav. 10 (2020), 2336-2351), we have investigated the reproducibility of graph measures calculated at the subject level based on resting state fMRI. We have investigated different processing pipelines and now we want to investigate if our selected pipeline is able to show differences among cognitively normal subjects (CN), patients with mild cognitive impairment (MCI) and patients with Alzheimer's disease (AD). More specifically we want to evaluate the relationship between graph measures and the cognitive assessments and we want to test if graph measures are predictive for cognitive decline. We will use a whole brain based parcellation as the nodes of the graph and we will use (partial) correlations between fMRI time series as measure of connectivity. We will apply our pipeline to determine the graph measures at the individual level and use standard statistical approaches to 1) test if there is a difference between graph measures between NC, MCI and AD; 2) determine the relation with cognitive assessments and 3) test if graph measures can predict cognitive decline.
