| Proposed Analysis: | This will be a longitudinal cohort study investigating the association between medication use and the development and progression of cognitive impairment and dementia. The main exposure will be medication use (including those used for the treatment and management of dementia and comorbidities). Outcomes will include various measures of cognition, including cognitive assessments, diagnoses, and biological markers (imaging and biospecimens). Covariates will include demographic, clinical, environmental and genetic factors. Cox regression models will be used to estimate hazard ratios and 95% confidence intervals for the association between predictors and incident MCI or dementia. Linear mixed effects models will be used to assess changes in cognitive outcomes in relation to medication use and other factors. |
