Proposed Analysis: Anatomical connectivity might reveal neurofibrillary differences of the axons prior to the detection of the neurodegeneration and symptoms in the gray matter (Braak and Del Tredici 2015). This will be consistent with the first specific aim in elucidating the connection mechanism of Alzheimer’s disease from the connectome point of view. I will collaborate with Associate professor Dr. Kesheng Wang, School of Nursing, Health Sciences Center, West Virginia University, Morgantown, WV, to study the imaging data in elucidating the mechanism of AD in the connections point of view. A novel method called fixel-based analysis (Raffelt et al 2015) will be adopted for statistical analysis of white matter integrity in greater detail exhibiting within-voxel microstructures for complex fiber crossings in brain. FA, fractional anisotropy, will be measured, a reflection of the strength of the connection. With the same approach, we will assess the vascular changes that are associated with the changes in long distance axonal connections.
