Proposed Analysis: The immune system plays essential roles in Alzheimer’s disease (AD). To explore the involvement of the key component, the cytokines, in the development of AD, we performed deep sequencing of 200 cytokine genes in case-control samples. We found that regulatory variants within several cytokines, e.g. EGF and IFNGR1, showed significant association with AD risk. Indeed, we do find the dysregulation of these cytokines in brain and blood tissues of AD patients. It is unclear whether the cytokines are altered in AD CSF, and whether the level of cytokines changes along disease development. We therefore apply for the biomarker data from ADNI, to explore the cytokine profiles at protein level. Subjects will be group to CN, MCI and AD, and sub-grouped to baseline and longitudinal stage, according to diagnosis. Proteomic, ELISA, and clinical data will be used, and differential protein abundant analysis will be performed to screening out candidate cytokines.
