Proposed Analysis: To date, genome-wide association studies have identified a number of SNPs significantly associated with Alzheimer’s disease, but these SNPs typically have a small effect, indicating that individual SNPs have limited predictive power. Some statistical techniques have developed to aggregate the effects of SNPs across the genome. A polygenic risk score (PRS), which is one of the techniques, is an estimate of an individual genetic liability to a disease calculated as a sum of effect allele counts weighted by corresponding genotype effect size. In the proposed study, we will investigate associations of AD-related polygenic risk score (AD-PRS) calculated by biological pathways based on the GWAS summary statistics and clinical diagnosis of AD and CSF biomarkers.
