Proposed Analysis: | Our proposed analysis plan for the requested ADNI data will focus on the development of novel accelerated failure time survival models to assess associations between biomarkers and the joint risk of Alzheimer’s Disease (AD) and death. First, we plan to develop a novel accelerated failure time survival model with percentile-varying effects that accommodates left-truncation and interval censoring, to capture new insights into the effects of risk factors on Alzheimer’s onset. Second, we plan to develop a novel Bayesian group-structured variable selection method for accelerated failure time survival models with left-truncation and interval censoring, to identify genetic risk factors for Alzheimer’s onset. Third, we plan to develop a novel Bayesian framework for the joint analysis of semi-competing risks and longitudinal data, to assess the impact of longitudinal biomarkers jointly on Alzheimer’s onset and mortality. Using the proposed high-dimensional methodology, we intend to model association between genetic markers and the joint outcomes of AD and death.
