Proposed Analysis: Growth differentiation factor 15 (GDF15) is a cytokine produced in response to inflammatory stressors and related to cognitive decline and some imaging markers of neurodegeneration. However, the possible mechanisms underlying this association are unknown. GDF15 may be simply a risk marker of dementia rather than a component in the causal path. Alternatively, GDF15 may be causally related to dementia. Thus, we aims to use Mendelian randomization approach to examine whether genetically predicted levels of GDF15 were associated to cognition, and imaging markers of Alzheimer's disease, cerebral small vessel disease and normal aging. Mendelian randomization analysis requires estimates of the effect sizes of both the association between the single SNPs and the exposure (i.e. GDF15 levels), and the associations between SNPs and the outcome (i.e. dementia). Specifically, we plan to apply for genetic data, study data, MRI data, and PET data to conduct a two-sample Mendelian randomization analysis. The SNPs associated with plasma GDF15 levels have been found in a previous GWAS study. Next,we used GWAS summary data form ADNI to obtain the effect size estimates of the association between SNPs and dementia as well as imaging markers.
