Proposed Analysis: | Our group (composed of two statisticans, an applied mathematician, and several graduate and undergraduate students ) is currently developing a statistical framework for spatial modeling in imaging genomics. Current interest lies in the development of methods for brain-wide genome-wide studies, where the associations between more than 100,000 SNPs and measures at more than 10,000 voxels of a 3D brain image are of interest. In addition to the multiple testing problem, the situation is made difficult by a lack of independence among tests, both over SNPs located in a particular gene, as well as over the voxels located in similar regions of the brain. In addition, the effect sizes of interest are likely weak, in particular if only a single voxel is considered. Approaches for analysis that have been proposed to date include massive univariate scans and various multi-locus methods that can be applied to examine joint associations separately at each voxel of a brain image, or at the very most, jointly over a small collection of brain summary measures. Explicit spatial models examining associations jointly across all voxels of an image have not been considered. Spatial models may prove useful in facilitating the borrowing of information across neighboring voxels leading to increased power and efficiency in the estimation of effect sizes. We intend to develop the statistical models and associated computational algorithms, and use data from ADNI to illustrate our methodology. |
