Proposed Analysis: | We are requesting individual level data from ADNI African American participants in order to generate AD GWAS summary statistics stratified by APOE genotype, age, and sex for meta analysis with results from the MVP dataset. We have conducted a GWAS in the Million Veterans Program African Americans using a dementia phenotype that combines individuals classified as AD and AD-related dementia (ADRD), noting that latter group includes predominantly AD cases. We will repeat these analyses stratified by APOE ε4 carriers vs. non-carriers, by sex, and by age at onset for cases and age at exam for controls. We will adjust for ancestry PCs, as well as for age and sex as is appropriate for each stratified analysis. All variants with imputation quality > 0.4 and MAF > 1% will be included. We will generate analogous results from the ADGC cohorts including ADNI, and meta-analyze these results with those from MVP, and perform follow-up bioinformatics analyses including pathway analysis and functional annotation using FUMA. Analyses will be conducted using methods suitable for related individuals where appropriate and utilize logistic or Firth logistic regression models implemented in PLINK or R. |
