Proposed Analysis: | Several studies have shown that there are patients with Alzheimer's disease (AD) who do not follow the existing Braak stage, raising doubts about the existence of subtypes of AD. The genetic mutations found in the existing case-control genome-wide association study (GWAS) may be common disease-related genetic mutations found regardless of AD subtypes, or may be a subtype causative genetic mutation found in a sufficiently large sample size. To discover significant new genetic mutations in samples with fewer sample sizes, it is necessary to defining subtypes of AD and performing GWAS with the subtypes. Therefore, I would like to define AD subtypes according to brain atrophy patterns and proceed with GWAS for each subtypes using genetic data, clinical information, MRI images and PET images from ADNI1/GO/2/3. MRI and PET Images will be preprocessed using Freesurfer. Next, I will adjust covariates such as age and gender for each brain region cortical volume or thickness extracted from Freesurfer then use Bayesian-based clustering methods. After dividing the AD subtypes, the GWAS between the case group and the control group for each subtype will be performed with PLINK. |
