Proposed Analysis: 80% OF Alzheimer's patients exhibit cerebrovascular pathology before the development of amyloid plaque burden, which can be detected early on in the disease. I hope to use the ADNI data set to investigate changes in genes for cerebrovascular inflammation such as vascular cell adhesion molecule-1, which is expressed at the blood brain barrier (BBB). I hope to correlate the changes in VCAM-1 gene expression in Alzheimer's patients to the extent of amyloid beta plaque deposition and glucose deprivation using Neuroimaging data from markers such as 18-FDG and Pib. This analysis could able to predict extent of amyloid beta deposition based on the extent of changes in expression of inflammatory markers expressed at the BBB. Additionally, we would like to investigate the changes in genes involved in pericyte maturation and functioning and correlate it with the changes in cerebrovascular inflammation and BBB integrity.
