Proposed Analysis: | Vascular risk factors, such as type 2 diabetes mellitus (T2DM), hypertension, and hyperlipidemia influence the vulnerability and progression of Alzheimer's disease (AD). However, the underlying molecular mechanisms remain elusive. DNA methylation is an important form of epigenetic modification, which plays a key role in regulating gene expression without altering the DNA sequence. DNA methylation changes occur in response to environmental factors (including the levels of blood sugar, blood pressure, and blood lipid) throughout whole life. The Alzheimer’s Disease Neuroimaging Initiative (ADNI) is an important data source available to the scientific community that greatly contributes to the understanding of AD. Therefore, we sought to use this resource to determine the DNA methylation profiles of T2DM, hypertension, and hyperlipidemia in the context of AD. This will reveal the epigenetic mechanisms of the involvement of T2DM, hypertension, and hyperlipidemia in AD, also point to potential drug targets for precision medicine.
