Proposed Analysis: | Evaluate epistasis in all pairwise interactions of genes for narrowly and broadly defined AD and ADRD at the transcription. We will apply newly developed TWIS analyses (currently in review: https://doi.org/10.1101/2022.08.16.504187) to identify gene-gene interactions using imputed expression that influence ADRD risk. We will genetically characterize the similarities and differences between narrowly defined AD and broadly defined ADRD and other dementias. Analyses will be performed using imputed expression in brain regions of key importance to ADRD risk (see Aim 1 above). Test enrichment of interaction associations of these a priori defined gene sets using E-TWIS, a method we recently developed, testing whether pathways and gene sets have stronger gene-gene interactions within them than expected by chance, thereby identifying key cell types, pathways and networks that underlie ADRD risk. Epistasis has never been systematically explored in ADRD risk, but TWIS has identified novel pathways and core genes integrating multiple signals in other complex traits. We propose an exhaustive, genome-wide gene-gene interaction study of ADRD, in which we will uncover new aspects of ADRD biology. |
