Proposed Analysis: The purpose of the analysis is to study the sequential ordering of CSF, MRI, and clinical biomarkers along the pathophysiological pathway of Alzheimer's disease. We propose a latent variable mixture model that quantifies disease progression by a dynamic latent metric. The latent metric accounts for individualized time-varying heterogeneity and is cluster-specific for identifying subgroups with systematically different risk profiles. With this analysis, we will account for the different roles of age in natural aging and pathophysiological process. In addition, Nudelman et al. (2014) discussed the association between cancer history and later age of AD onset. I am interested in investigating the association between baseline cancer history and AD progression using our mixture model.
