Proposed Analysis: | Hippocampal Morphometry: Analyze surface-based shape changes of the human hippocampus in those with Alzheimer's Disease (AD). Hippocampal volumes will be segmented from T1-weighted MR images. Volumes will then be processed through SlicerSALT (a shape analysis-focused fork of 3D Slicer) using spherical harmonics (SPHARM-PDM) to generate surface meshes of every hippocampus, complete with alignment and registration. Vector-based distances between every correspondent vertex will be calculated from each subject's hippocampus at multiple timepoints, then inputted into a statistical linear model paradigm alongside variables such as amyloid/tau deposition levels (SUVR values, etc.) and cognitive test scores. P-value "heat maps" of the hippocampal surface will show regions of shape change that are significantly correlated with AD diagnosis/status, cognitive test scores (multiple domains), amyloid/tau deposition levels, etc. Results will be used to inform and guide clinical approaches to diagnosis and identification of AD pathology at various stages of progression. |
